Androgenic activation, impairment of the monoaminergic system and altered behavior in zebrafish larvae exposed to environmental concentrations of fenitrothion

Abstract Fenitrothion is an organophosphorus insecticide usually found in aquatic ecosystems at concentrations the range of low ng/L. In this manuscript we show that 24 h exposure to environmental fenitrothion, from ng/L ?g/L, altered basal locomotor activity, visual-motor response and acoustic/vibrational escape zebrafish larvae. Furthermore, fenitrothion expression gap43a, gfap, atp2b1a, mbp exhibited a significant non-monotonic concentration-response relationship. Once determined were neurotoxic for larvae, computational analysis identified potential protein targets compound. Some predictions, including interactions with acetylcholinesterase, monoamine-oxidases androgen receptor (AR), experimentally validated. Binding AR was most suitable candidate molecular initiating event, as indicated by both up-regulation cyp19a1b sult2st3 relationship between observed responses. Finally, when integrity monoaminergic system evaluated, levels L-DOPA, DOPAC, HVA 5-HIAA found, well slc18a2 lowest fenitrothion. These data strongly suggest commonly present risk fish communities.
